Glutton for punishment and fast healer do not seem to stack that I've noticed, but both are good individually. I find efficient harvest better for getting more resources quickly. There aren't really enough 'rare' resources for careful harvest to be worth it, besides maybe goldstone from obsidian. Well, Corrupted Vitality is good for the first 9 levels. The Grotesque Excrescence perk heals more than the Fierce Vitality. Other than that, not worth losing the Fast Healer perk.
